Robotic Platform Advances Forensic DNA Testing

Forensic DNA Extraction Method Validated on Eppendorf epMotion™ 5075-TMX

Chromosomal Laboratories, Inc is excited to announce that is has successfully pioneered the development and validation of an automated method for using the Prepfiler extraction kit on the Eppendorf epMotion™ 5075-TMX platform.

The marriage of the latest robotic technology with the recently released Prepfiler™ DNA extraction kit from Applied Biosystems optimizes extraction of forensic samples containing trace DNA while concurrently increasing laboratory throughput and reducing manual manipulation of critical specimens..

Unlike other robotic systems on the market, the Eppendorf epMotion™ 5075-TMX provides an open platform to create an unlimited number of single tube method formats. President Nicole Bolin, who is pioneering the methods for Chromosomal Laboratories, stated that, “The epBlue software is user-friendly and scripts can be written easily and quickly. The software offers a “simulation” run, which allows the user to observe the robot movements (scripts) on the computer monitor prior to running the script on real samples. This simulation allows the script to be reviewed without wasting reagents and consumables.”

The Prepfiler™ system has been refined to incorporate uniquely structured magnetic particles that yield much greater surface area. This has enabled much greater DNA capture and when combined with a new wash buffer, allows for a vastly improved removal of most inhibitors that negatively affect DNA recovery.

This new extraction has proven highly efficacious for DNA recovery from body fluid stains, “touch DNA”, hair roots, fingernails and tissue. When combined with the epMotion robotic system, extraction time for a 96 well plate has been reduced to two hours. Mrs. Bolin, who will be presenting her validation studies later this year at the 21st International Symposium on Human Identification adds that, “In addition to the improved efficiency of the epMotion™ 5075, Prepfiler™ has enabled our company to consolidate three of our previous extraction techniques into just one, while limiting the use of two carcinogenic chemicals from our lab.”
